Synthesis and cancer cell growth inhibition effects of acetogenin analogs bearing ethylene glycol units for enhancing the water solubility
2020, TetrahedronCitation Excerpt :Moreover, Wu and co-workers reported that acetogenin analogs bearing an ethylene glycol unit instead of the THF-ring moiety also exhibited growth inhibitory activity against human cancer cell lines;[17] hence, we concluded that the ethylene glycol unit of 2b may behave like another THF moiety. Similarities between the antitumor mechanisms of compounds can be predicted by COMPARE analysis,[18] where the Pearson correlation coefficients are calculated to measure similarities between various combinations of compounds. Thus, a fingerprint, or a pattern of log GI50 values of the various compounds across the JFCR39 cell lines, was employed for comparison, which revealed that analogs 2a–2c exhibited similar fingerprints to that of lead compound 1 (Table 3, r = 0.668, 0.615, and 0.658, respectively).

Our first attempted reaction with Et3N in the presence of (1R,2S)-N-methylephedrine (NME) in toluene gave propargyl alcohol 8a in low yield with moderate diastereoselectivity (entry 1).12 The diastereoselectivity was determined by analyzing the 1H NMR spectra of acetate 9 after acetylation with Ac2O in pyridine.

In order to develop lead compounds for new anticancer agents, our group has synthesized various analogues of annonaceous acetogenins [2–4]. Annonaceous acetogenins [5], which are polyketides isolated from Annona plants growing in tropical and subtropical regions, potently inhibit NADH ubiquinone oxidoreductase (complex I) in the mitochondrial electron transport system [6].
