Abstract

ラット根尖病変の治癒におけるトランスフォーミンググロースファクター(TGF)-β1発現細胞の動態について免疫組織学的に検索した。実験的に誘発されたラット根尖病変へ抗菌剤(オフロキサシン)を根管貼薬した結果,TGF-β1を発現したマクロファージの著明な増加とその後の急激な減少が認められた。また,根尖周囲では著しい骨形成が観察された。一方,生理食塩液貼薬群では少数のTGF-β1を発現したマクロファージの浸潤が認められた。しかし,骨面では多数の骨芽細胞が配列するにも関わらず,根尖周囲の消失した骨組織は回復されなかった。加えて,生理食塩液貼薬群では多数のIL-1αおよびIL-1βを発現したマクロファージが継続して病変中に認められたのに対して,オフロキサシン貼薬群ではIL-1αおよびIL-1βの発現は著しく減少した。以上のことより,TGF-β1の発現期間および発現の程度が治癒の進行に影響し,TGF-β1を発現したマクロファージの一時的な増加が,根尖性歯周炎治癒過程における新生骨形成の活性化において,一つの重要な役割を果たしている可能性が示唆された。
The aim of this study was to examine the behavior of cells expressing transforming growth factor (TGF)-β1 during the healing process of rat periapical lesions. After applying the antibacterial agent (ofloxacin) to the experimentally induced periapical lesions, a significant increase followed by a drastic decrease in TGF-β1-expressing macrophages and considerable bone formation was seen around the root apex. On the other hand, topical application of physiological saline resulted in only small numbers of TGF-β1-expressing macrophages. However, lost bone tissue within the vicinity of the root apex was not easily recovered despite there being numerous osteoblasts on the bone surfaces. In addition, expression of IL-1α and IL-1β was considerably decreased in ofloxacin-treated rats, whereas there remained numerous IL-1α and IL-1β-expressing macrophages in the lesions of physiological saline-treated rats. These results suggest that the time and extent of TGF-β1 expression may influence the course of healing, and that a transient increase of TGF-β1-expressing macrophages may play an important role in the activation of new bone formation during the healing process of apical periodontitis.
